Insights from active surface theory for engineering division in synthetic cells
Building a synthetic cell capable of division is a decisive step for bottom-up synthetic biology on its way towards a minimal living system. Reconstitution of division modules has made substantial progress in assembling contractile rings
placing them at mid-cell and initiating ingression
and this modular route has the clear advantage of being controllable and experimentally tractable. It has
however
repeatedly stalled at the step that matters most: sustaining ingression down to a thin neck and completing membrane scission. What is missing is a mesoscale criterion for deciding
before an experiment is run
whether a deformation will keep amplifying or relax back
which leaves parameter optimization largely empirical. Active surface theory offers a class of testable stability criteria of this kind. It treats the cell surface as a thin active layer driven by chemical reactions
which flows in plane and bends out of plane
and closes the description with force and torque balance
constitutive laws constrained by Onsager reciprocal relations
and a mechanochemical feedback loop coupling regulator concentration
active tension
surface flow and geometry. The central proposition advanced here is that active surface theory is not a replacement for the modular reconstitution route but a mesoscale organizing principle added to it: given the constraints set by membrane area
volume
dissipation channels and scission mechanism
it asks whether the reconstituted active layer can enter the dynamical regime in which a division ring forms and ingression is sustained. Ring formation and ingression are accordingly framed as a mechanochemical instability controlled jointly by orientational organization
dissipation length scales and component dynamics. Two design implications are emphasized: within the nematic active surface model discussed here
the degree of filament orientational order
rather than the absolute magnitude of contractile stress
governs whether the equatorial mode becomes unstable
and excessively strong isotropic contractility may further suppress that mode by destroying nematic order; and the ratio of vesicle radius to hydrodynamic screening length constitutes a dynamical scale-matching condition independent of the intrinsic curvature of the filaments. Recent FtsZ-cytoFtsN experiments are consistent with this picture at the mesoscale level: higher filament order and longer polymer lifetime favour large-scale constriction. Because cytoFtsN acts mainly through charge-mediated bundling and kinetic stabilization
however
this cannot yet be regarded as direct validation of the active alignment coefficient. Specific models further predict multistability and hysteresis during ingression
which can be tested by bidirectional sweeps of a control parameter. Because the framework describes continuous deformation only
complete division additionally requires an adequate membrane area reserve and a dedicated scission mechanism. Suggestions are made for parameter calibration and for closing a quantitative loop between theory and reconstitution.
